      光遗传学是一种通过联合使用光学技术和重组DNA 技术实现对细胞行为进行精确控制的变革性研究手段,在神经科学及其它相关领域已展示出巨大的应用价值。目前国内医学院校关于光遗传学实验技术原理及应用的课程教学远没有跟上快速发展的神经科学大趋势。本文对国内医学院校开展针对本科生的光遗传学实验技术原理与应用教学的必要性、主要内容、教学难点、以及教学实施过程中应注意的理论和实践问题进行了初步分析,特别强调了应重点讲解的技术原理,包括其何以实现对特定的神经通路进行选择性干预,何以分别实现对胞细胞的兴奋或抑制效应的双向控制,以及开展光遗传学实验的主要过程。最后,我们对近期在授课过程中的体会进行了初步总结,以供相关院校基础医学实验教学参考,并推动国内医学院校本科生光遗传学实验教学的开展。

      Optogenetics is a transformative research method combining optical technologies with recombinant DNA technologies to realize the precise control of cellular behaviors. Optogenetics has shown wide application in neuroscience and other related research fields. A lot of important research progresses have been made in neuroscience in recent years due to the usage of this technology. However, course teaching of the principle and application of the optogenetics experimental technology for undergraduates in domestic medical colleges has not kept up with the rapid development of neuroscience. In view of the importance of optogenetics experimental technology in many research fields, we have made a preliminary discussion here on the teaching of principles and applications of optogenetics experimental technology for medical undergraduates.Optogenetics is a transformative research method combining optical technologies with recombinant DNA technologies to realize the precise control of cellular behaviors. Optogenetics has shown wide application in neuroscience and other related research fields. However, course teaching of the principle and application of the optogenetics experimental technology for undergraduates in domestic medical colleges has not kept up with the rapid development of neuroscience. Here we have made a preliminary analysis on the necessity, main contents and difficult points in teaching medical undergraduates the principles and applications of optogenetics experimental technology. We focused on the theory and practice teaching about this technique and emphasized on the most important technological principles, including how to achieve selective intervention of specific neural pathways, how to realize the two-way control, i.e,. excitation or inhibition, of neurons, and the main process of the experiment. Finally, we have summarized our recent experience in the teaching process, to provide reference for basic medical experiments teaching in related medical colleges, and to promote optogenetics experimental teaching in domestic medical colleges.
